14-3-3 proteins in apoptosis BJMBR
Rosenquist,M..
The once obscure members of the 14-3-3 protein family play significant roles in the determination of cell fate. By inhibiting the pro-apoptotic BAD (Bcl-2-antagonist of cell death) and the transcription factor FKHRL-1, 14-3-3 displays important anti-apoptotic characteristics. To date, five points of interaction of 14-3-3 with the apoptotic machinery have been identified. How these interactions are regulated still remains a mystery.

A DNA enzyme targeting Egr-1 inhibits rat vascular smooth muscle cell proliferation by down-regulation of cyclin D1 and TGF-β1 BJMBR
Wu,Y.; Han,W.; Liu,G.-N..
We have demonstrated that a synthetic DNA enzyme targeting early growth response factor-1 (Egr-1) can inhibit neointimal hyperplasia following vascular injury. However, the detailed mechanism of this inhibition is not known. Thus, the objective of the present study was to further investigate potential inhibitory mechanisms. Catalytic DNA (ED5) and scrambled control DNA enzyme (ED5SCR) were synthesized and transfected into primary cultures of rat vascular smooth muscle cells (VSMCs). VSMC proliferation and DNA synthesis were analyzed by the MTT method and BrdU staining, respectively. Egr-1, TGF-β1, p53, p21, Bax, and cyclin D1 expression was detected by RT-PCR and Western blot. Apoptosis and cell cycle assays were performed by FACS. Green fluorescence could...

A herpes-like virus infects a non-ostreid bivalve species: virus replication in Ruditapes philippinarum larvae ArchiMer
Renault, Tristan; Lipart, Cecile; Arzul, Isabelle.
Sporadic high mortalities were reported in June 1997 among hatchery-reared larval Manila clam Ruditapes philippinarum in a French commercial hatchery. Cellular abnormalities were observed in semi-thin sections in affected animals. Transmission electron microscopy revealed the presence of herpes-like virus particles in larvae. This is the first description of a herpes-like virus infection in larval R. philippinarum, a non-ostreid bivalve species. Virus particles were similar to other herpes-like viruses described from different oyster species with respect to ultrastructure and morphogenesis. Electron microscopic examination also demonstrated cells with condensed chromatin and extensive perinuclear fragmentation of chromatin. Like viruses infecting oysters,...

Adiponectin is protective against endoplasmic reticulum stress-induced apoptosis of endothelial cells in sepsis BJMBR
Hou,Yun; Wang,Xi Feng; Lang,Zhi Qiang; Jin,Yin Chuan; Fu,Jia Rong; Xv,Xiao Min; Sun,Shi Tian; Xin,Xin; Zhang,Lian Shuang.
Endoplasmic reticulum (ER) stress is a critical molecular mechanism involved in the pathogenesis of sepsis. Hence, strategies for alleviating this stress may be essential for preventing cardiovascular injuries under sepsis. Adiponectin is secreted by adipocytes and its levels are decreased in sepsis. The purpose of this study was to investigate the protective effects of adiponectin treatment on endothelial cells and its mechanism. Male Wistar rats underwent cecal ligation and puncture (CLP) before being treated with adiponectin (72 and 120 μg/kg). The levels of malondialdehyde (MDA) in plasma, histological structure, and apoptosis of endothelial cells were evaluated. In vitro, human umbilical vein endothelial cells (HUVECs) were treated with adiponectin at...

Advanced glycation end products (AGEs) and their receptor (RAGE) induce apoptosis of periodontal ligament fibroblasts BJMBR
Li,D.X.; Deng,T.Z.; Lv,J.; Ke,J..
Diabetics have an increased prevalence of periodontitis, and diabetes is one of the causative factors of severe periodontitis. Apoptosis is thought to be involved in this pathogenic relationship. The aim of this study was to investigate apoptosis in human periodontal ligament (PDL) fibroblasts induced by advanced glycation end products (AGEs) and their receptor (RAGE). We examined the roles of apoptosis, AGEs, and RAGE during periodontitis in diabetes mellitus using cultured PDL fibroblasts that were treated by AGE-modified bovine serum albumin (AGE-BSA), bovine serum albumin (BSA) alone, or given no treatment (control). Microscopy and real-time quantitative PCR indicated that PDL fibroblasts treated with AGE-BSA were deformed and expressed higher levels...

AGEING INDUCES APOPTOSIS AND INCREASES HSP70 STRESS PROTEIN IN THE EPIDIDYMIS OF Octodon degus International Journal of Morphology
Bustos-Obregón,Eduardo; Esponda,Pedro.
Apoptosis has been largely analyzed in the testis. Nevertheless, the epididymis has been scarcely studied. We analyzed the number of apoptotic cells in the different regions (caput, corpus and cauda) of the epididymis of the South American rodent Octodon degus both young and senile. Apoptosis was identified using the TUNEL method which detects in situ DNA fragmentation. Apoptosis was detected in the principal cells of the epididymal epithelium. The caput epididymis was the region more affected. The caput of young animals showed that 0.32/1000 cells were apoptotic in contrast to 5.1/1000 of senile animals. Also in the cauda epididymis apoptosis is increased with age, appearing 0.14/1000 and 3.9/1000 in young and senile animals, respectively. On the other...

AKT inhibitor suppresses hyperthermia-induced Ndrg2 phosphorylation in gastric cancer cells BJMBR
Tao,Yurong; Guo,Yan; Liu,Wenchao; Zhang,Jian; Li,Xia; Shen,Lan; Ru,Yi; Xue,Yan; Zheng,Jin; Liu,Xinping; Zhang,Jing; Yao,Libo.
Hyperthermia is one of the most effective adjuvant treatments for various cancers with few side effects. However, the underlying molecular mechanisms still are not known. N-myc downstream-regulated gene 2 (NDRG2), a tumor suppressor, has been shown to be involved in diverse cellular stresses including hypoxia, lipotoxicity, etc. In addition, Ndrg2 has been reported to be related to progression of gastric cancer. In the current study, our data showed that the apoptosis rate of MKN28 cells increased relatively rapidly to 13.4% by 24 h after treatment with hyperthermia (42°C for 1 h) compared to 5.1% in control cells (P < 0.05). Nevertheless, there was no obvious change in the expression level of total Ndrg2 during this process. Further investigation...

Alterations in gene expression profiles correlated with cisplatin cytotoxicity in the glioma U343 cell line Genet. Mol. Biol.
Carminati,Patricia Oliveira; Mello,Stephano Spano; Fachin,Ana Lucia; Junta,Cristina Moraes; Sandrin-Garcia,Paula; Carlotti,Carlos Gilberto; Donadi,Eduardo Antonio; Passos,Geraldo Aleixo Silva; Sakamoto-Hojo,Elza Tiemi.
Gliomas are the most common tumors in the central nervous system, the average survival time of patients with glioblastoma multiforme being about 1 year from diagnosis, in spite of harsh therapy. Aiming to study the transcriptional profiles displayed by glioma cells undergoing cisplatin treatment, gene expression analysis was performed by the cDNA microarray method. Cell survival and apoptosis induction following treatment were also evaluated. Drug concentrations of 12.5 to 300 μM caused a pronounced reduction in cell survival rates five days after treatment, whereas concentrations higher than 25 μM were effective in reducing the survival rates to ~1%. However, the maximum apoptosis frequency was 20.4% for 25 μM cisplatin in cells analyzed at 72 h,...

Antifungal and antitumor models of bioactive protective peptides Anais da ABC (AABC)
Rodrigues,Elaine G.; Dobroff,Andrey S.; Taborda,Carlos P.; Travassos,Luiz R..
Peptides are remarkably reactive molecules produced by a great variety of species and able to display a number of functions in uni-and multicellular organisms as mediators, agonists and regulating substances. Some of them exert cytotoxic effects on cells other than those that produced them, and may have a role in controlling subpopulations and protecting certain species or cell types. Presently, we focus on antifungal and antitumor peptides and discuss a few models in which specific sequences and structures exerted direct inhibitory effects or stimulated a protective immune response. The killer peptide, deduced from an antiidiotypic antibody, with several antimicrobial activities and other Ig-derived peptides with cytotoxic activities including antitumor...

Antioxidant dietary deficiency induces caspase activation in chick skeletal muscle cells BJMBR
Nunes,V.A.; Gozzo,A.J.; Juliano,M.A.; Cerqueira César,M.; Sampaio,M.U.; Sampaio,C.A.M.; Araújo,M.S..
Apoptosis and necrosis are two distinct forms of cell death that can occur in response to different agents and stress conditions. In order to verify if the oxidative stress induced by dietary selenium and vitamin E deficiencies can lead muscle cells to apoptosis, one-day-old chicks were reared using diets differing in their vitamin E (0 or 10 IU/kg) and selenium (0 or 0.15 ppm) supplementation. Chick skeletal muscle tissue was obtained from 28-day-old animals and used to verify apoptosis occurrence based on caspase activity detection and DNA fragmentation. Antioxidant deficiency significantly increased caspase-like activity assessed by the hydrolysis of fluorogenic peptide substrates (Abz-peptidyl-EDDnp) at lambdaexc = 320 nm and lambdaem = 420 nm....

The glaucophyte Cyanophora paradoxa (Cp) was chemically investigated to identify pigments efficiently inhibiting malignant melanoma, mammary carcinoma and lung adenocarcinoma cells growth. Cp water and ethanol extracts significantly inhibited the growth of the three cancer cell lines in vitro, at 100 mu g.mL(-1). Flash chromatography of the Cp ethanol extract, devoid of c-phycocyanin and allophycocyanin, enabled the collection of eight fractions, four of which strongly inhibited cancer cells growth at 100 mu g.mL(-1). Particularly, two fractions inhibited more than 90% of the melanoma cells growth, one inducing apoptosis in the three cancer cells lines. The detailed analysis of Cp pigment composition resulted in the discrimination of 17 molecules, ten of...
